细胞毒性化疗在肿瘤的综合治疗中仍被广泛应用,临床实践已明确消化道毒性及骨髓抑制仍是滞碍患者接收化疗的原因。中医药可以减轻化疗毒副反应,却缺乏系统深入的机制研究。本团队前期实验表明,六君安胃颗粒对肠道中疣微菌门和梭杆菌门相对丰度的降低有一定帮助。基于此,本课题拟从体内及体外实验分别进行六君安胃序贯芪菟二至颗粒减轻化疗所致消化道毒性或骨髓抑制的疗效,分析比较中药干预后小鼠粪便肠道菌群分布,菌群功能基因的改变及菌群代谢产物的改变,Wnt/β-catenin信号通路上蛋白表达,基因转录情况及干细胞的变化,探明六君安胃颗粒/芪菟二至颗粒-肠道菌群/菌群代谢物-Wnt/β-catenin信号通路介导的肠道干细胞/骨髓造血干细胞再生-肠黏膜损伤修复/骨髓造血的复杂网络关系。探究中医先后天理论与Wnt/β-catenin信号通路介导的干细胞再生的关系,从分子生物学的角度解释中医健脾补肾理论的科学内涵。

Chemotherapy is still widely used in the comprehensive treatment of tumors. Clinical practice has shown that gastrointestinal and bone marrow toxicity are still the reasons that hinder patients from receiving chemotherapy. Traditional Chinese medicine can alleviate the side effects of chemotherapy, but there is no systematic and in-depth mechanism research. Previous experiments of our team showed that Liujun'anwei Granule was helpful to decrease the relative abundance of microflora verrucosae and clostridium in intestinal tract. Based on this, this study intends to conduct in vivo and in vitro experiments on the effect of Liujun'anwei Granule sequential Qituerzhi Granule on reducing gastrointestinal and bone marrow toxicity induced by chemotherapy, and to analyze and compare the distribution of intestinal flora in mice feces after the intervention of traditional Chinese medicine, the changes of functional genes and metabolites of flora, the expression of Wnt/β-catenin signaling pathway protein, gene transcription and stem cell regeneration.To explore the complex network relationship of intestinal stem cells/bone marrow hematopoietic stem cells regeneration-intestinal mucosal injury repair/bone marrow hematopoiesis mediated by Liujun'anwei/Qiqierzhi Granule-intestinal flora/bacterial metabolite-Wnt/β-catenin signaling pathway. To explore the relationship between congenital theory of TCM and stem cell regeneration mediated by Wnt/β-catenin signaling pathway, and to explain the scientific connotation of the TCM theory from the perspective of molecular biology.
