Up to now the molecular mechanisms that regulate memory reconsolidation are still largely unknown. As the critical downstream negative regulator of G protein coupled receptor (GPCR), β-arrestin attenuates GPCR-mediated signal transduction and induce receptor internalization in an agonist-dependent manner. Due to the great distribution in the brain, the functions of β-arrestin in central nerves system attract more attention now. We have been focusing on the research of the neurological roles of β-arrestin for several years. We found that the negative regulation of PKA in amygdale by β-arrestin 2 / phosphodiesterase-4 complex was critical for fear conditioning. Based on our previous research interests and results, we try to demonstrate the role of β-arrestin in the reconsolidation of object recognition memory and spatial memory, then try to find out the new function of β-arrestin in the central nerves system. In this research, the β-arrestin global knockout mice and conditional knockout mice are used to perform the object recognition memory tasks. The lentivirus and associated adenovirus system to knockdown or expression β-arrestin are also introduced in this study to test the role of β-arrestin in memory reconsolidation. Then the neural plasticity and molecular mechanism regulated by β-arrestin in memory reconsolidation will be focused. This research will supply some neurobiological clues for the memory dynamic storage dependent neurocircuits and molecular mechanism.
记忆稳定需要经历巩固和再巩固过程,关于记忆再巩固的分子机制一直是研究热点。记忆再巩固机制假说中一个重要观点认为β-肾上腺素受体通过下游G蛋白介导的PKA信号通路参与记忆再巩固的形成。β-肾上腺素受体下游负性调控分子β-arrestin介导受体内吞并终止G蛋白依赖信号的传递,而β-arrestin也能独立发挥信号传递的功能,但其在中枢神经系统的作用尚不清楚。本项目在我们以往工作的基础上,采用β-arrestin基因敲除小鼠,结合药理学手段,聚焦于β-arrestin参与记忆再巩固的调控作用及调控的分子机制的研究。我们发现β1选择性拮抗剂倍他洛尔能抑制小鼠物体辨别记忆的再巩固,而G蛋白偏向性β-肾上腺素受体拮抗剂卡维地洛不仅不影响物体辨别记忆的再巩固,还能促进恐惧记忆的再巩固。卡维地洛不仅能抑制G蛋白信号通路,还能激活β-arrestin2依赖的ERK信号通路,提示非G蛋白依赖的β-肾上腺素受体信号通路在记忆再巩固中的作用。进一步研究结果显示β-arrestin2敲除小鼠表现出物体辨别记忆再巩固受损的行为表型,提示了β-arrestin2在记忆再巩固中的重要作用。研究还发现在物体辨别记忆再巩固过程中,内嗅皮层β-arrestin2依赖的ERK信号通路显著激活。β-arrestin2在内嗅皮层的表达能恢复β-arrestin2敲除小鼠的ERK信号通路的激活和物体辨别记忆再巩固受损的行为表型,该作用能被倍他洛尔抑制,但不能被卡维地洛抑制。证明了β-肾上腺素受体通过β-arrestin2偏向性受体信号通路参与物体辨别记忆的再巩固。因此,本项目研究提出β-肾上腺素通过β-arrestin2依赖的信号通路而不是G蛋白信号通路介导了记忆再巩固的新假说(Proc Natl Acad Sci U S A, 2015)。在此基础上,我们的工作还发现了β-肾上腺素受体信号通路参与调控非条件性刺激诱导条件性恐惧记忆的再巩固(Front Neural Circuits., 2017; Front Behav Neurosci. 2017),以及Rac1激活在短时程和长时程条件性恐惧记忆形成中的作用(Front Mol Neurosci., 2015)。期间共发表论文9篇,其中在PNAS等刊物上发表本基金标注的SCI论文5篇,我们的结果提示了记忆再巩固的分子机制,为记忆相关疾病提供重要治疗靶点。
