植株衰老后叶片仍然保持绿色而不变黄的特性称为持绿性。持绿性蔬菜品种对于以叶片为产品器官的大白菜具有特殊重要意义。申请人前期以大白菜小孢子培养获得的DH系‘FT’为试材,采用萌动种子EMS诱变处理方法,创制出1份持绿突变体nym1,其全生育期植株保持绿色不变黄,其它性状与野生型基本一致。遗传分析证明了nym1的持绿性呈单基因隐性遗传,遂命名为Brnym1。利用BSR-Seq方法,已将Brnym1定位于A10染色体的2.2Mb区域。本项目拟进一步开展Brnym1基因克隆与鉴定研究:利用F2群体精细定位突变基因,筛选确定候选基因;通过转基因技术互补验证候选基因的功能;利用qRT-PCR、启动子活性分析及亚细胞定位技术摸清突变基因的表达特性。同时,探索持绿性突变引起的突变体在抗衰老和抗逆性上的变异。研究结果可以揭示持绿突变基因的功能与作用,为更好的开发利用这一珍贵的遗传资源奠定。
英文摘要
Stay-green refers to a trait whereby plants retain leaf greenness during senescence. The stay-green varieties have special significance for Chinese cabbage whose leaves were as product organs. In previous research, an excellent Chinese cabbage DH line ‘FT’ was employed as the plant material, whose germinated seeds were treated by EMS to create mutants. A stay-green mutant, ‘nym1’, was obtained. The leaves of nym1 remained green throughout the whole growth period, even in the aging stage. Moreover, the other characters of mutant were consistent with the wild-type ‘FT’. Genetic analysis indicated that the stay-green trait of nym1 was controlled by a simple recessive nuclear gene, designated as Brnym1. Using BSR-Seq technology, Brnym1 was located within a distance of 2.2Mb, on the chromosome A10. In this project, the study on cloning and functional identification of Brnym1 will be performed. Fine mapping of Brnym1 will be conducted using the F2 separation population and the candidate gene will be identified. The function of the candidate gene will be validated by transgenic technology. The expression characteristics of the Brnym1 will be analyzed by real-time quantitative PCR, promoter activity analysis and subcellular localization technology. At the same time, the mutation of anti-aging and stress resistance in nym1 which caused by stay-green mutation will be explored. These research results contribute to reveal the function of the Brnym1, and lay the foundation for developing and utilizing the precious genetic resources of stay-green.
