家蚕质型多角体病毒(BmCPV)是危害家蚕的重要病原之一,长链非编码RNA(lncRNA)参与转录及转录后调控等多个生命活动,在病毒感染与宿主免疫中发挥重要调控作用,是近年来生命科学领域的研究热点。本项目拟以家蚕应答BmCPV感染明显上调表达的lncRNA-LINC20486为对象,研究lncRNA对BmCPV感染、复制与致病的调控作用及其分子机制。研究内容包括LINC20486的基本特性、亚细胞定位以及在家蚕各组织中的表达谱;筛选与LINC20486互作的靶分子,探究LINC20486对靶基因/蛋白表达的调控及其调控作用机制;探究LINC20486及其对靶分子的调控作用对BmCPV感染、复制与致病的影响及其分子机制。研究结果有助于解析BmCPV在家蚕体内复制及感染致病的分子机理,为家蚕质型多角体病的诊断与防治提供新的思路,并为家蚕抗病分子育种提供新的靶点。
英文摘要
Bombyx mori cypovirus (BmCPV) is one of the most important pathogens causing severe disease to silkworm. Long non-coding RNA (lncRNA) is involved in various biological processes, including transcriptional and post-transcriptional regulation. LncRNA plays importantly regulatory roles in virus infection and host immune response, which has been a hotspot in the territory of life sciences research in recent years. In the current study, we focus a BmCPV-induced lncRNA-LINC20486 to analyzed the molecular mechanism of BmCPV infection and replication. Major issues to be studied include: the basic characteristics and subcellular localization of LINC20486 and its expression in various tissues of silkworm; the regulation of LINC20486 on target molecules and its molecular mechanism; the effect and mechanism of LINC20486 in regulating BmCPV infection, replication and pathogenicity. The results will be helpful to understand the molecular mechanism of BmCPV replication and infection in silkworm, and provide a new direction for the diagnosis and control of cytoplasmic polyhedrosis. Besides, the key lncRNA and genes identified in the study could be used as target molecules for genome editing to breed virus-resistant silkworm varieties.
