尿酸代谢紊乱可引发高尿酸血症、痛风和肾功能衰竭。研究表明人体内产生的尿酸有1/3通过肠道菌群分解代谢,但是哪些菌群在发挥作用以及深层次的机制仍不清楚。我们前期研究发现肠道菌群与高尿酸血症有一定的关联,且哈萨克族尿酸水平远低于其他民族,但该民族膳食中嘌呤水平远高于其他民族。于是提出“肠道菌群在尿酸代谢中发挥重要作用”这一假说。为验证此假设 :1)我们拟采用高通量序列测定方法了解哈萨克族不同人群中肠道菌群的分类、群落结构、系统进化及遗传多样性;2)采用体内实验对具有降解尿酸活性细菌进行筛选和驯化、菌种鉴定和相关指标检测,探索可能与哈萨克族高嘌呤低尿酸特性有关微生物的作用机制,3)通过体外实验对筛选出的具有高效降解尿酸菌株进行功能验证,期望找到可能影响尿酸代谢的靶标菌株。本研究从微生态角度探讨尿酸代谢紊乱发病机制,为尿酸代谢紊乱的早期预警以及应用特异菌株的个性化治疗和药物研制提供科学依据。
英文摘要
Uric acid metabolic disorder can cause hyperuricemia, gout and renal failure. Studies have shown that 1/3 of the uric acid produced in the human body is metabolized by intestinal flora, but it is still unclear which flora are playing a role and the underlying mechanism. Previous studies have found that intestinal flora is associated with hyperuricemia, and the level of uric acid in Kazakh is much lower than that in other ethnic groups, but the level of purine in the diet of this ethnic group is much higher than ethnic groups. so we put forward the hypothesis“intestinal flora plays an important role in uric acid metabolism”. To verify this hypothesis, we intend to use high-throughput sequencing method to understand the classification, community structure, phylogenetic evolution and genetic diversity of intestinal flora in different populations of Kazakh people; 2) The vivo experiments will be used to screen and domesticate bacteria with uric acid-degrading activity, identify bacteria species and detect related indicators to explore microorganisms that may be related to the characteristics of high purine and low uric acid in Kazakh people., 3) Found some strains with have high uric acid degradation in vitro experiments, in order to find target strains that may affect uric acid metabolism. To explore the pathogenesis of uric acid metabolic disorder from the perspective of microecology, and provide scientific basis for early warning of uric acid metabolic disorder, personalized treatment of specific strains and drug development in the future.
