随着全球气候变暖,了解环境和生物化学因素是如何调节木本植物内休眠过程,对农业生产至关重要。目前,寻找与木本植物内休眠相关的基因是一个活跃的领域。FLC基因是春化调控途径的关键基因,MaMADS33为桑树中FLC-like基因。前期的研究表明,MaMADS33通过抑制MaFT基因的表达,从而抑制内休眠芽的萌发。为了寻找更多的证据证明MaMADS33基因参与桑树的内休眠过程,本课题基于前期研究的两条线索进行解析:1、MaMADS33与桑树冬季积累蛋白WAP18互作;2、MaMADS33基因受ABA的诱导。首先明确MaMADS33和WAP18的互作参与到桑树的内休眠过程,逐步解析ABA调控途径与MaMADS33在内休眠过程中的关系,再利用桑树转基因方法进一步分析MaMADS33在内休眠过程中的生物学意义。本课题的实施将丰富木本植物内休眠过程调控的分子机理,为人工调控桑树内休眠提供理论和实践基础。
英文摘要
As the global warming, it is crucial for agricultural production that how environmental and biochemical factors affect the regulation of endodormancy in woody plants. Now, it is an active field relating to genes involving woody endodormancy. FLC plays important role in vernalization. MaMADS33 is one of the FLC-like genes in mulberry. Previous studies showed that MaMADS33 suppressed the expression of MaFT to inhabit the sprout of dormancy buds. To find more evidence to verify the importance of MaMADS33 in endodormancy, this subject base on two clues of previous research, 1. MaMADS33 protein could interact with winter accumulated protein 18; 2. MaMADS33 gene was induced by ABA. The first step is to confer the involvement of the interaction of MaMADS33 and WAP19 in endodormancy. And then, the relationship between ABA pathway and MaMADS33 in endodormancy will be analazed. And finally, the mulberry transgenetic will be used to uncover the biology signification of MaMADS33 in endodormancy. This project enriches the molecular mechanism of endodormancy regulation in woody plants. It also provides theoretical and practical basis for artificial regulation of endodormancy in mulberry.
