英文摘要
Lichens are highly successful and stable mutualistic organisms between fungi and either or both green algae and cyanobacteria. However, attempts in the laboratory to re-create a lichen using only these two partners were never successful. Recently, a third symbiont that builds the outer cortex of the lichen was discovered in several lichen genera, which belongs to Basidiomycete yeast. Although Cladonia is one of the earliest described genera due to its great abundance in nature and prominent morphological features, its morphogenesis is poorly understood. This species-rich genus (about 500 accepted species) is ecologically important due to its wide occurrence and abundant secondary metabolites. The preliminary research mainly based on DNA barcoding genes has shown that the endolichenic fungi associated with cladoniiform species comprise not only a high degree of taxonomic diversity, but also genetic traits and functional diversity that is both unique relative to the endophytes in other biomes, and potential use for antifungal activity. In the project, we are going to identify fungal components in cladoniiform lichen thalli and understand their prevalence in natural populations using hologenomic data, to localize and simultaneously visualize different fungal bionts in lichen thalli by FISH and CLSM, to isolate and culture both the lichenized and endolichenic fungi, to study the physiological function of Basidiomycete yeast in cladoniiform lichen and to re-create the whole cladoniiform lichen, and to reconstruct the multilocus (ITS,partial LSU and MitSSU etc.) phylogeny for the different endolichenic fungal partner which will provide an evolutionary framework for both the ascomycetes and the basidiomycetes embedded in the lichen thalli and will lead to the description of a few new species. Meanwhile, the culture-based and culture-free approaches will be developed to unveil the species diversity, spatial heterogeneity, environmental sensitivity, and host preference of endolichenic fungi associated with some important cladoniiform species at both local and regional scales.
地衣是以真菌-藻类共生为主的多成分微生物群落,是生物间互惠影响的典型范例。了解与地衣相关的微生物群落的作用仍然是一项科学挑战。当真菌入侵并破坏藻类细胞时,光合作用伙伴与真菌伙伴之间的关系就从互惠关系转变为寄生关系。然而,地衣内生担子菌酵母却构成了地衣共生的另一部分,或可称其为内生建成真菌。至于这些担子菌酵母是否会带来生态和生理方面的益处,目前仍存在争议,需要进一步研究。石蕊属地衣因其分布广泛及形态复杂,而凸显生态重要性,但对其形态建成仍知之甚少。在石蕊属的一些种类的外皮层中,发现有担子菌酵母。.通过本项目的研究,我们利用源于真菌扩增子高通量测序PacBio获得的全基因组数据识别出石蕊属中重要种类地衣体中大量的各种类型的真菌组分,并对它们在自然居群中的分布情况及丰富程度有了更深入的了解。这些真菌组分包括子囊菌门12纲1984种、担子菌门12纲884种、以及其他8个真菌门中的96个种级分类单元;其中属于担子菌酵母类群的有囊担子菌纲38个、银耳纲191个。通过荧光原位杂交和共聚焦激光扫描显微镜定位到3种石蕊属地衣地衣体中担子菌酵母组分在皮层中的位置。分离得到地衣型真菌、地衣内生真菌及共生藻类156种792株,隶属于16纲28目42科64属;其中的12种102株为担子菌酵母;探讨了内嵌的担子菌酵母在重建完整地衣体时的作用,但未能成功实现地衣体的人工重建。通过培养和非培养方法相结合的全面分析显示石蕊属地衣种类的内生真菌没有明显的宿主偏好性及地域偏好性。基于奥变期的新型分子钟模型估算柄锈菌亚门中各种来源的担子菌酵母类群的分化时间,构建其系统发育演化框架,作出拟建立2个新目、1个新科、2个新属、并恢复地衣担子菌属的分类处理。发现石蕊属地衣4个新种、3个中国新记录种。申报了食用菌藻组合增效有利于人类健康的发明专利。
