Cell assemblies are essential for multiple brain functions, but damage to nodes within these assemblies can cause functional failure. It is hypothesized that activating remaining cell assemblies is essential to restore lost function. Activating pathways that bypass the damaged routes may be a potential method to achieve this. Electrical stimulation is a potential method to activate the bypassing pathway, given its potential to modify the synaptic connection between stimulated neurons.The research aimed to establish an in vitro platform to study how electrical stimulation can encode functions by modifying connections between neuronal assemblies. Two main results were obtained: establishing an in vitro neuronal system to study information transformation and evaluating the system to acquire information given by electrical stimulation programs.The developed system, composed of neocortical and hippocampal networks, demonstrated cross-frequency coupling properties via burst activities, which indicates the usage of the system in studying information transformation between multi-component neuronal systems, as published in Frontiers in Neuroscience.Using this system, a stimulation program called "time interval training" was developed to regulate the spiking relations between neurons, allowing for the adjustment of the burst activities propagation time between the two sub-cultures. Live imaging methods were then used to identify the participation of different neurons in burst events, providing valuable insight into the mechanism of the designed system for information possession.
