Preventive Effects of Prostaglandin Inhibitors and Antioxidants on the Experimental Uveitis.

1)介绍：为了研究抗炎药物如阿司匹林对实验性葡萄膜炎的预防作用，本实验观察了阿司匹林和类固醇对内毒素诱导的家兔实验性葡萄膜炎的影响，其作用可能与内毒素诱导的葡萄膜炎中的内毒素合成有关。注射后16小时，将1 ml内毒素（20 μ g）玻璃体内注射至兔眼，摘除眼并分离虹膜。作为炎症的指标，测定虹膜中的髓过氧化物酶（MPO）。结果：玻璃体内注射生理盐水组虹膜组织中未发现MPO活性，内毒素组虹膜组织中MPO活性明显升高，达2.9 × 10 ~（-6）U/ml，而对照组虹膜组织中MPO活性明显降低，达2.9 × 10 ~（-6）U/ml，差异有显著性（P &lt; 0.05）<-2>。注射阿司匹林50 mg/kg以上组和泼尼松龙1 mg/kg以上组的虹膜髓过氧化物酶（MPO）活性抑制约15%。结论：阿司匹林对内毒素性葡萄膜炎有一定的预防作用。这一结果表明，洋地黄素可能参与了内毒素诱导的葡萄膜炎。其他结果：内毒素处理的眼睛的虹膜中的脂质过氧化没有增加。MPO活性在S抗原诱导的葡萄膜炎的虹膜现在正在调查中。

1)Introduction : In order to study the preventive effect of anti-inflammatory drugs such as aspirin, a prostagalandin inhibitor, on the experimentally induced uveitis, the effects of aspirin vs steroid on endotoxin-induced experimental uveitis in the rabbits which may be related to the prostaglandins synthesis were studied.2)Methods : O. 1ml of endotoxin(20, mug)was intravitreally injected to the rabbit eye 16 hrs after the injection, the eye was enucleated and the iris was separated. As a indicator of the inflammation, myeloperoxidase(MPO)in the iris was determined. Before the intravitreal injections of endotoxin, aspirin DI-lysine(25mg/kg, 50mg/kg, 100mg/kg)or prednisolone(0.5mg/kg, lmg/kg, 2mg/kg)were performed.3)Results : No activity of MPO in the iris of the eyes which received the intravitreal injection of normal saline solution was-found. MPO activity of the iris of the eyes treated with endotoxin increased highly as 2.9x10^<-2>U/Mg protein.MPO activity of the iris was inhibited by about 15% in the groups treated with the injections of more than 50mg/kg of aspirin and in that of more than lmg/kg of prednisolone. However, there was no significant Difference in the both groups.4)Conclusion : The preventive effect of aspirin on the endotoxin-induced uveitis could be demonstrated. This result suggests that prostaglandins may be involved in the endotoxin induced uveitis.Other results : Lipid peroxidation did not increase in the iris of the endotoxin- treated eye. MPO activity in the iris of S-antigen-induced uveitis is now under investigation.
