Les végétaux demeurent une source incontournable de molécules bioactives. En réponse à des stimuli externes comme la prédation, des agents pathogènes ou des stress environnementaux, les plantes produisent des molécules de défense qui possèdent des propriétés biologiques uniques. Même si ces molécules sont très attrayantes pour le développement de médicaments, leur étude et utilisation sont souvent limitées par leur faible solubilité et leur faible biodisponibilité. Une approche prometteuse pour surmonter ces limitations consiste à modifier le squelette d'un produit naturel. Appelée dérivatisation, cette stratégie a déjà fait ses preuves pour améliorer les propriétés physico-chimiques et le profil pharmacologique des produits naturels. Dans ce projet, nous nous intéressons particulièrement aux triterpènes pentacycliques de la famille des lupanes qui proviennent de l'écorce du bouleau blanc Betula papyrifera. L'objectif du projet est donc de dérivatiser une série de lupanes bioactifs du bouleau blanc par fonctionnalisation et expansion de cycle pour modifier leurs propriétés physico-chimiques et permettre leur utilisation dans différents tests d'activité biologique afin d'investiguer les propriétés biologiques de ces nouvelles structures. Nous espérons ainsi développer de nouvelles méthodes de synthèse, générer des structures moléculaires innovatrices, améliorer les propriétés pharmacologiques des lupanes et découvrir de nouvelles bioactivités pour ces produits. À terme, ce projet permettra de valoriser l'écorce de bouleau, une ressource du Canada ainsi qu'un rejet de l'industrie forestière en produits à des fins industrielles, vétérinaires et pharmaceutiques.
