The project aims to answer the question by which responsive nanocarrier-drug conjugates with different physico-chemical properties penetrate into and act in the skin as a function of the physical and biochemical environment in 3D skin models, human, and murine skin tissue by using advanced time-resolved fluorescence methods. Developments include multiparameter and multiphoton fluorescence lifetime imaging (FLIM, FLIM-FRET, PLIM) for the tomographic and physiological analysis of inflamed skin. A combination of single molecule total internal reflection (TIRF) microscopy with FLIM for single molecule detection in the hair follicle planned.
