先行研究(Hayano T, Matsui H et al. PLoS One. 2016.)である前立腺癌家系を用いたエクソーム解析の結果から、BRCA1/2遺伝子バリアントが複数同定された。今回BRCA2遺伝子の17遺伝子変異に着目し、日本人家族性前立腺癌における変異頻度を明らかにし、家族性、散在性前立腺癌の関連検討することで、臨床データからCRPC(castration-resistant prostate cancer)および前立腺癌死症例とHSPC(Hormone-Sensitive Prostate Cancer)の関連解析を行い致死的な遺伝子変異の同定を目的とする。対象は日本人家族性前立腺癌242名、日本人散発性前立腺癌198名および対照として年齢をマッチさせた前立腺肥大症（非前立腺癌）142例とした。これまでにダイレクトシークエンスとTaqMan SNP Genotyping Assayゲノタイピング結果の妥当性の確認をした後、実際の検体を用いたゲノタイピングを行い、全体の9割り程度のゲノタイピングが終了している（Table 1）。アレル頻度は極めて低いバリアントが多いようである。今後、ゲノタイピング結果から、家族性前立腺癌、散発性前立腺癌のBRCA2　17バリアントとの関連、臨床データ（家系内の共有、予後、去勢抵抗性前立腺癌か否か等）による統計学的解析を進めていく。
