抗血管内皮細胞抗体（AECA）の対応抗原であるheat shock cognate 71 kDa protein (HC70)およびtropomyosin alpha-4 chain (TPM4) に対する自己抗体を検出するためにELISAをおこなった。血清中の自己抗体は、好酸球性多発血管炎性肉芽腫症（EGPA）の日本人患者10人および日本人健常者7人で測定された。HSC70に対する自己抗体は、EGPAの30％で検出され、健常人では検出されなかった。一方、TPM4に対する自己抗体はEGPAの10％で検出され、健常人では検出されなかった。さらに、EGPAで抗HSC70抗体もしくは抗TPM4抗体が検出された患者では、全症例でANCAが陰性であった。喘息患者由来ヒト肺微小血管内皮細胞（HLMVEC-AS）からの抽出蛋白とEGPAの日本人患者血清を用いたウエスタンブロッティング(WB)では、ANCA陽性患者に比べ陰性患者で多数のAECAが検出された。特に、約40、50、100および150kDaのタンパク質に対する自己抗体が測定した全5症例のANCA陰性EGPAで検出され、ANCA陽性EGPAでは検出されなかった。現在、プロテオーム解析を行いANCA陰性EGPAもしくはEGPAに特異的なAECAの対応抗原候補蛋白を検出・同定している。

All 5 cases of ANCA were detected by 150kDa antibody test, ANCA EGPA test and ANCA antibody test. Now, we need to analyze the AECA antigen candidate protein of the ANCA sex EGPA protein specific to the AECA antigen.
