The endosome represents a switchboard where membrane proteins and lipids are sorted towards different destinations within the pre- and the postsynaptic compartments and thereby maintaining their homeostasis. To understand the basic neuronal function of the endosome-associated and Parkinson¿s disease-related protein RME8 (human ortholog DNAJC13) we will analyze its role in synaptic vesicle and neurotransmitter recycling. We will establish a novel conditional mouse model to investigate in vivo the importance of the RME8 in maintaining neuronal circuits. In addition, we will uncover the role of RM8 in endosome-derived exosome release and their uptake by neighboring cells as exosomes are believed to contribute to the prion-like propagation of protein misfolding in neurodegenerative disease.
