In this research, an HPLC multiwavelength detection system was developed which can measure the absorption spectra of components in addition to their retention time. The developed system was also applied to actual analysis of biological fluids such as human urine to evaluate its effectiveness.Hardware controlled by several microcomputers and needed control software for the HPLC multiwavelength detection system were developed. It can measure and record spectra over a range of 200nm to 800nm. A data processing system was specially designed for effective and efficient processing of the large amounts of data obtained by multiwavelength detection. Data processing programs were developed which can extract useful information from data obtained by a single analysis of a sample. Examples of these programs are 3-dimensional displays of chromatograms and comparison of chromatograms of different samples using computer graphics. Human urine samples from patients with various diseasis and from healthy controls were analyzed. The results show that some peaks, which may appear to represent a singele component by using a monowavelength absorbance detector, are found to actually represent several components when they are analyzed using this multiwavelength detection system. In the 3D chromatograms of urine samples from some diseased groups, there are some peak patterns which are not observed in the chromatograms of urine samples from healthy groups. These results show the effectiveness of the developed system in the field of biological fluid analysis. Statistical analysis and multivariate analysis of peak heights in the chromatograms were tested to develop methods to detect these differences by automatic computer analysis. The results show that the patterns of amount changes of several components in urine samples are different between diseased groups and healthy groups. This point must be furhter investigated in future research.
