项目摘要

EBウイルスのDNAはヒト・インターロイキン10(hIL-10)遺伝子と高い相同性を有する遺伝子BCRF1を保有する。さらにEBV感染によりhIL-10の発現が誘導されることも知られる。BCRF1やhIL-10はB細胞性成長因子として、また抗原特異的キラーT細胞の抑制因子として働くことによりEBV感染Bリンパ球増殖や生体における免疫回避に働いていることが予想される。そこで各種EBV陽性腫瘍細胞株を用いてEBV潜伏感染遺伝子発現と併せて両遺伝子の発現を調べ、その免疫生物学的活性を検討した。EBV不死化Bリンパ細胞株では全6種類のEBV特異的核抗原EBNAと3種類の膜抗原LMPの発現が見られたが、RT-PCR法によりhIL-10の発現が認められたがBCRF1遺伝子発現は認められなかった。一方、EBV陽性バ-キットリンパ腫細胞株Akataでは潜伏感染遺伝子はEBNA1のみが発現しており、hIL-10は発現していないがBCRF1遺伝子発現が認められた。Akata細胞でのBCER1遺伝子発現は抗免疫グロブリン抗体によりEBV活性化誘導により増強した。健康人末梢リンパ球とEBVトランスフォーム細胞を混合培養してEBV特異的キラーT細胞を誘導する系にAkata細胞培養上清を添加するとキラーT細胞活性の低下が認められたことから、BCRF1遺伝子が生体におけるEBVの免疫回避に関与する可能性が示唆された。

EB-10 DNA fragment has high homology with BCRF1 fragment. The development of hIL-10 in EBV infection was induced by the virus. BCRF1 and hIL-10 are antigen-specific inhibitors of B-cell growth factors and antigen-specific inhibitors of T-cell growth factors. A variety of EBV-positive tumor cell lines were used to detect EBV latent infection, modulate EBV latent infection, and investigate their immunobiological activity. The expression of all six EBV-specific nuclear antigens EBNA and three membrane antigens LMP in EBV inactivated B cell lines was detected by RT-PCR and the expression of hIL-10 was detected by RT-PCR. In one case, EBV-positive cell line Akata was found to have latent infection with EBNA1, hIL-10 and BCRF1. Akata cell BCER1 gene expression increases in response to induction of EBV activation by anti-immune antibodies Healthy human peripheral blood cells and EBV cells were cultured in a mixture and EBV-specific T cells were induced in Akata cell culture supernatants. The decrease in T cell activity was recognized as a possible link between EBV immune avoidance and BCRF1 gene production in vivo.
