1)ストレスによるHPA軸の活動を抑制するアンドロジェンの作用の検討昨年度、シバヤギにおいて、アンドロジェンが輸送に伴う血中コルチゾル(Co)濃度の上昇を軽減させることを示した。本年度は、この現象に関わるアンドロジェンの作用を解明する一環として、副腎皮質刺激ホルモン放出ホルモン(CRH)の脳室投与による血中Co濃度の変化に及ぼすジヒドロテストステロン(DHT)の効果を検討した。去勢したオスシバヤギの皮下にDHTあるいはコレステロール(Cho)を詰めたカプセルを留置し、第三脳室に生理食塩水あるいはヒツジCRHを注入した。血中Co濃度は、DHT群、Cho群ともにCRHの脳室注入により有意に上昇したが、その上昇の程度はDHT群の方が有意に低かった。このことから、アンドロジェンの主な作用部位は中枢ではなく、下垂体あるいは副腎皮質であることが示唆された。2)輸送による摂食行動の変化における雌雄差および性腺ホルモンの影響輸送後の摂食行動の変化と、それに関わる雌雄差と性腺ホルモンの影響を検討した。1時間の輸送を行ない、その直後から6時間の摂食量を測定した。輸送後の摂食量は、メスでは対照と差がなかったが、オスでは輸送により減少していた。また、去勢オスにCho、DHTあるいはエストラジオールを詰めたカプセルを留置して行った実験では、DHT留置群でのみ輸送により摂食量が減少していた。輸送後の摂食量の減少が何に起因するかは今のところ不明であるが、血中Co濃度における反応で得られた結果とは逆に、アンドロジェンがその効果を促進していることが示唆された。3)輸送が視床下部のc-fosタンパク発現に及ぼす影響輸送により、視床下部に発現するc-fosの分布を検討した。去勢オスを用い、1時間の輸送後、抗ヒトc-fos抗体を用いて免疫染色をした。その結果、輸送により、HPA軸の活動に主要な役割を果たす室傍核にc-fosの発現が認められた。しかし、交感神経系を調節している視床下部腹内側核にはc-fosの発現はみられなかった。これらの発現に、性腺ホルモンがどのような影響を与えるかは今後の課題である。
