A small caliber vascular prosthesis remains unsatisfactory in regard with the long term patency rate. A hybrid vascular prosthesis has been investigated for achieving the better patency in the way of combining the living cells with a synthetic graft. We studied the endothelial cells (ECs) seeding to the ultrafine polyester fiber grafts and developed the filtration seeding method in order to utilize a large number of cells. The in vitro and in vivo study revealed that the ECs grew up around the grafts with time and maintain the continuous monolayer of the ECs three months after implantation. However, at the anastomotic sites, the ECs monolayer was formed on the thick subendothelial tissue which continued to the thick intima of the host artery. The ECs seeding only may not be enough to prevent the graft failure. The intimal hyperplasia may be the major target for achieving the better patency of the small caliber vascular prosthesis in the following research.
