ラットおける5alpha-reductaseの遺伝子発現をRT-PCRによって作成したcADNA probe を用いて検討した。ラットにおいてはnorthern blot analysisによって、5alpha-reductaseのシグナルを確認することができ、このシグナルはtestosterone分泌抑制処置によって増加傾向を示すことから、testostroneによるdown-regulationの可能性が示唆された。このようにアンドロゲン環境そのものによって5alpha-reductaseの遺伝子発現は調節を受けることが考えられる。ヒト前立腺癌でのnorthern blot analysisでは、我々がRT-PCR法によって作成したcADNA probeによってはシグナルを明らかにすることが困難なため、海外より入手したヒト5alpha-reductaseのcADNA probeによって確認を行っている。これによって、この原因がprobeに由来するものか、あるいは、transcriptの量的な減少に由来しているのかを推測することができるものと思われる。ヒト前立腺癌リンパ節転移由来のLNCaP細胞に発言している5alpha-reductasの酵素力学的検討から、この発言が通常の前立腺に見られる酵素と質的に異なることをKm値の相異から推定しているが、さらにpHに対する反応性の違いからもこの点は裏付けられるにいたった。
