14年度に続き、広島大学医学部歯学部附属病院歯周診療科を受診し、口腔内臨床所見、レントゲン所見および歯周病関連細菌に対する血清抗体価検査の結果から早期発症型歯周病と診断された患者を選び、早期発症型歯周病患者のデータベースを作成した。本年度は14年度の30名につづき70名の患者を登録し、合計100名の登録患者が得られた。登録患者には好中球機能検査を実施した。検査は好中球の貪食能および走化能について調べた。14年度と同様に、好中球貪食能が正常者に比べて低下している患者はいなかった。好中球走化能を合成FMLPを用いてジグモイドチャンバー法によって調べた結果、正常者に比べて有意に低下している患者2名をスクリーニングした。2名の患者については再検査を行い好中球走化能が低下していることを確認した。14年度、15年度に好中球走化能低下を明らかに認め、本研究に同意の得られた3名の患者から末梢血を採取して、好中球を分離した。比較対象として歯肉に炎症が認められず、4mm以上の歯周ポケットが存在しない成人健常者3名の好中球も分離した。分離した好中球を合成FMLPによって刺激した後に、総蛋白質を得た。得られた蛋白質を2次元電気泳動によって分離展開し、成人健常者の好中球の電気泳動パターンとの比較をコンピュータ画像処理によってディファレンシャル分析した。その結果、早期発症型歯周病と診断され好中球走化能低下を認める患者の好中球に発現の増加するタンパク質スポット群、発現の低下するタンパク質スポット群を得た。
