项目摘要

1. 歯周ポケット内細菌のDNAマーカーの分離とDNAの解析1) 歯周ポケット細菌からの直接Diffrential display歯周ポケットから採取した細菌を直接Differential displayを行うと,安定した結果を得ることが出来なかった.2) 歯周ポケット細菌の前培養とDifferential display歯周ポケットから採取した細菌を初めにプレートに播種し増菌し,性状の異なるコロニーを単離しDifferential displayを行った.前培養,増菌という作業を加えることによって,安定した結果を得ることが出来た.3) 標準菌株のDifferential displayPorphyromonas gingivalis,Actinobacillus actinomycetemcomitans,Prevotella intermediaなどの歯周病関連細菌の標準菌株をDifferential displayを行った結果,それぞれに特徴的なパターンを表示することが可能であった.2. 歯周病病態と細菌DNAマーカー1) 歯周炎の病態として,正常,早期発症型歯周炎,成人性歯周炎の患者からポケット内細菌を採取し,前述した前培養,増菌作業を行った後にDifferential displayを行った.研究計画に前培養,増菌作業が加わったことから,Differential displayだけで病態と関連する細菌DNAマーカーを同定することは出来ない事が明らかになった.2) 歯周炎の病態が異なる,正常,早期発症型歯周炎,成人性歯周炎の患者から分離したポケット内細菌を採取し,プレートに播種し増菌し,性状の異なるコロニーを単離しDifferential displayを行った結果,病態ごとに異なる細菌をDifferential display法によってそのパターンの違いを表示することが可能なことが明らかになった.

In the pre-culture, the standard strains of Differential displayPorphyromonas gingivalis,Actinobacillus actinomycetemcomitans,Prevotella intermedia bacteria were tested, and the results of diazepam tests were negative. 3) the standard strains of Differential displayPorphyromonas gingivalis,Actinobacillus actinomycetemcomitans,Prevotella intermedia bacteria were tested by standard strains of bacteria with weekly disease, and the standard strains of bacteria were tested by Differential display. 2. The pathogens of DNA disease were isolated from patients with periodontitis, normal, early symptomatic periodontitis and adult periodontitis. The bacteria were collected from the patients with chronic periodontitis, and the bacteria were cultured before operation and Differential display was performed after operation. Study program pre-clinical culture, bacterial operation plus laboratory screening, Differential display screening of pathogenic bacteria, DNA isolates of pathogenic bacteria. 2) Periarthritis of periodontitis, normal, early symptomatic periodontitis, adult periodontitis patients, isolation of pathogenic bacteria, isolation of bacteria from patients with human periodontitis, isolation of pathogenic bacteria from patients with human periodontitis. The trait was isolated from the Differential display. The results of the Differential display method indicate that it is possible to indicate that it is possible to detect the disease.
