Molecular cloning of the genes for DNA repair in higher plants.

以水稻、二倍体烟草和大豆为材料，分别构建了直接文库、扩增文库、双扩增文库和两种消减文库。以DNA修复基因高度保守区的几种寡核苷酸为杂交探针，通过克隆杂交筛选阳性克隆。通过将它们导入修复缺陷型大肠杆菌和酿酒酵母中进行表达，获得了高等植物光解酶基因克隆。

"Direct" cDNA library, "amplified" cDNA library, "double amplified" cDNA library, and 2 types of "subtracted" cDNA library were constructed from mRNA of tissues of rice, diploid tobacco, and soybean. With some kinds of oligonucletides derived from highly conservative regions of DNA repair genes as hybridization probes, positive chones were screened by clony hybridization. By introducing and expressing them in repair defficient lines of E.coli and S.cereviside, the clones encoding photolyase of the higher plant were isolated.
