A STUDY FOR HUMAN INTERLEUKIN BETA PRODUCTION AND PROCESSING SYSTEM

我们对表皮IL-1及其受体系统进行了化学和生物化学研究。IL-1 β前体或成熟型特异性抗体在正常表皮中检测到proIL-1 β，但未检测到成熟IL-1 β。银屑病表皮减少，但含有造血细胞衍生的成熟IL-1 β。除角质形成细胞外，这些造血细胞均含有IL-1 β转化酶。表皮表达信号转导型IL-1受体1，非信号转导型IL-1受体2几乎不表达。器官培养的正常皮肤和银屑病皮损表皮上调诱饵IL-1受体的表达：2型受体，但不是1型受体。正常表皮表达的IL-1受体拮抗剂在银屑病皮损中耗尽。IL-1在炎症性皮肤病中表达增强，但IL-1的过度表达也受到多种天然拮抗系统的严格控制：受体拮抗剂和诱饵受体的过度表达。

We investigated epidermal IL-1 and IL-1 receptor system immunohistochemically and biochemically. Pro or mature form IL-1 beta specific antibody detected proIL-1 beta but not mature IL-1 beta in normal epidermis. Psoriatic epidermis decreased but contained hematopoietic cell derived mature IL-1 beta. These hematopoietic cells but nit keratinocyte contained IL-1 beta converting enzyme. Epidermis express signal transducable IL-1 receptor type 1 and non-signal transducable IL-1 receptor type 2 scarcely. Organ cultured normal skin and psoriatic lesional epidermis up-regulated expression of decoy IL-1 receptors : type 2 receptors but not type 1 receptors. The IL-1 receptor antagonist normally expressed on epedermis was exhausted in psoriatic lesions. The IL-1 expression was augmented in the inflammatory skin diseases but the over-expression of IL-1s were also strictly controlled multiple natural antagonistic systems : receptor antagonist and decoy receptor over expression.
