プログラムフリーザ-による凍結保存方法の再現性を向上させるため、予備実験として、以下の実験を行った。試料として入手したブタ心臓より大動脈弁および肺動脈弁を採取し、大動脈弁および肺動脈弁は採取後ただちに凍結保存液に移した。これをprogram freezerにより凍結し、液体窒素液層内に保存し、凍結後、1週間、1か月、3か月後に解凍した。試料は、トリパンブルー染色による線維芽細胞生存性の定量的評価を行い、凍結期間と細胞生存性の関連を検討した。また、ブタ心臓より大動脈弁および肺動脈弁を採取し、DTC199液体培地凍結保存液内に同種弁を浸し、上記と同様の方法を用いて1か月間凍結保存した(T群)。同様に採取した大動脈弁および肺動脈弁をDMSOを除いたCell-Bankerで6時間処理した後、DMSOを含むCell-Bankerを加え、上記の方法で1か月間保存した(B群)。急速に解凍し、試料の線維芽細胞生存性の定量的評価を行い、凍結保存液の相異と線維芽細胞生存性の関連を検討した。現在実験は継続中であり、凍結保存を終了し、組織生存性の検討を行っている。現在までに得られた組織生存性は以下のとおりである。B群:control67.3%、24時間後60.1%、1週間後(採取後凍結)63.9%、1か月後(採取後凍結)56.7%、1週間後(24時間後凍結)51.8%、1か月後(24時間後凍結)53.6%。T群:contro64.7%、24時間後67.1%、1週間後(採取後凍結)49.3%、1か月後(採取後凍結)65.3%、1週間後(24時間後凍結)63.9%、1か月後(24時間後凍結)56.5%。
