6週齢のウイスター系雌性ラット20匹を10匹ずつの骨粗鬆症群ならびに対照群に分けた.骨粗鬆症群は両側の卵巣を摘出して,低カルシウム食(Ca:0.02%)と蒸留水を与えることによって骨粗鬆症を発症させた.対照群に対しては偽手術を行って,通常食(Ca:1.15%)と蒸留水を与えた.観察期間は術後12および32週(18および38週齢)とし,屠殺の10日前にテトラサイクリン(体重100gあたり5.0mg)を,3日前にカルセイン(体重100gあたり0.8mg)を投与した.各観察期間毎に各群の5匹ずつを屠殺して,臼歯部口蓋組織を採取し,Villanuevaの骨染色を施した後,MMA樹脂に包理し,頬舌的に約50μmの未脱灰研磨標本を作製した.落射蛍光顕微鏡を用いて,病理組織学的に観察するとともに,二次元画像解析装置を用いて単位骨量,分画形成面率,分画吸収面率および石灰化速度を計測した.これらの結果とを併せて,骨粗鬆症が硬口蓋骨組織に及ぼす影響に関して検討を加えた.その結果,骨粗鬆症群では対照群に比べて,両観察期間を通じて骨量の明らかな減少がみられ,骨表面には破骨細胞による骨吸収と骨芽細胞による骨添加が生じている部位が多く認められた.単位骨量は対照群に比べて減少し,分画形成面率,分画吸収面率および石灰化速度は対照群に比べて増加した.以上の結果から,本研究で得られた骨粗鬆症の硬口蓋骨組織は,骨吸収と骨添加を増加させる高代謝回転型の変化を示すことが明らかになった.

The results of this study were discussed in detail, including the effects of ossosis on hard skull bone tissue and bone marrow. As a result, the bone mass of the osteoporosis group was reduced during the observation period, and the osteoclasts on the bone surface were absorbed and the bone buds were added. The unit mass decreases in contrast to the group, the percentage of surface area formed, the percentage of surface area absorbed and the rate of calcification increase in contrast to the group. The results of this study showed that the bone resorption and bone addition in the hard flap of osteoporosis were increased, and the hypermetabolic metabolism was also increased.
