本研究では、トポロジカルな構造情報のみに注目した構造類似性の定量的評価のためのアルゴリズムを考案するとともに対応するシステムの開発を目的とした。本研究では、先に筆者らが提案した部分構造列挙とその対応する部分グラフのグラフ特性量に基づく特徴づけによって生成されるフラグメントスペクトル表現をもとに化学構造のトポロジカルなプロフィールを記述した。各部分構造の特徴づけには構成原子の質量数の和を用いた。従って、ここでのフラグメントスペクトル表現は質量スペクトル様の表現として記述される。類似性の評価に際してはこれらのスペクトルを多次元パターンベクトルで表し、各パターン間のユークリッド距離に基づく類似度を定義した。また、これらの処理に対応するプログラムを開発するとともに一連のプロセスのシステム化を図った。作成したシステムをもとに、42種の向精神薬を用いてこれらの構造類似性解析を試みたところ、研究者の視覚的直感ときわめて良好な対応を有する類似性評価の結果を得ることができた。また、本法を化合物データベースの構造類似性検索の問題に応用し、合わせてその有用性を確認した。
