The role of Foxolin GH-mediated life span was analyzed by comparing life span in the mice with GHR-/-and GHR-/-Foxo1-/-. Increase of body weight in Ghr-/-Foxo1+/-mice was much more remarkable than in GHR-/-mice, indicating that Foxo1 may play an important role in the signaling cascade of IGF system under the GH deficient condition.Secondly, the role of Foxo1 on glucose metabolism was examined. Blood glucose profile in mice with Ghr-/-Foxo1+/-at the age of 2,4 and 6 months was significantly lower than in Ghr-/-mice. Plasma insulin level significantly decreased in male mice with Ghr-/-Foxo1+/-than Ghr-/-mice. This was not observed in female mice. These findings suggest that Ghr-/-Foxo1+/-mice manifests increased insulin sensitivity. However, sex difference was present in increased insulin sensitivity. It is interesting to know from now how body weight and insulin sensitivity change with advancing age and how is a life span.
