Vertebrate craniofacial morphology originates from cranial neural crest cells. The neural crest cells move into the maxillary and mandibular regions. After the cell migrations, the mesenchymal cells interact with tissues in their local environment and signals from the forebrain and the surface epithelium.The LIM homeodomain (Lhx) genes are necessary for the differentiation of central nervous system neurons. L3/Lhx8, a member of the LIM homeobox domain family of transcription factors is expressed in the craniofacial regions and essential for the formation of secondary palate.We describe the identification and expression pattern of L3 /Lhx8 during embryogenesis of the chick. The protein sequence of L3/Lhx8 chick EST clone was most homologous to mouse L3/Lhx8. Its amino acid identity was more than 90%. RT-PCR analysis during facial development revealed L3/Lhx8 mRNA in chicken maxillary process was detected at stage 19HH and then the expression gradually increased from stage 27 to 31HH. Frozen sections demonstrated restricted expression of L3/Lhx8 in the maxillary mesenchyme and the apposed corners of the frontonasal mass but never in the overlaying epithelium. When removed the maxillary epithelium at stage 18HH, the embryo developed defect of maxillary bone with cleft lip. Our data suggests that an epithelial-mesenchymal interaction induced L3/Lhx8 expression, which may be responsible for development and formation of the upper jaw.
