It is well known that cyclooxygenase-2(COX-2) induces prostaglandin E2 (PGE2) synthesis and facilitates angiogenesis. Because synthesis of vascular endothelial growth factor (VEGF) is regulated by PGE2, COX-2 also might induce VEGF synthesis. We have investigated the expression of COX-2 mRNA in oral squamous cell carcinoma (SCC) cells, KA, NA, NI, KO and UE, and adenocarcinoma cells of salivary grand, HSG and HSY by RT-PCR. Any cells except UE expressed COX-2 mRNA. Nonsteroidal anti inflammatory drugs (NSAIDs), which inhibit cyclooxygenase enzyme activity, suppressed the growth of oral SCC cells expressing COX-2 mRNA. However, NSAIDs showed no effect on the growth UE cells lacking the expression of COX-2 mRNA. These findings suggest that NSAIDS, COX-2-Inhibitor inhibits angiogenesis by suppression of VEGF synthesis as well as proliferation
