アトピ-性皮膚炎(AD)患者、および正常人の末梢血よりFicollーHypaque法により単離した単核球を、RPMI1640にPhytohemoagglutinin(PHA),Interleukinー2(ILー2),Interleukinー4(ILー4)を添加して培養した。FcεRII陽性細胞は、PHA,ILー4添加培養で増加した。ILー2は単独では、FcεRII陽性細胞を増加させなかったが、PHAで刺激した後ILー2を加えると、FcεRII陽性細胞は増加した。特異的誘導(培養後のFcεRII陽性細胞%ー培養前のFcεRII陽性細胞%)は、ILーあるいはPHA添加培養では、約5ー10%でその約半数はT細胞であった。PHA+ILー2添加では、約5%であった。特異的誘導は、AD患者単核球と正常人単核球で、有意の差がなかった。上記培養中に、インタ-フェロン(IFN)γ,ーαおよびプロスタグランディンE_2(PGE_2)を添加して、レセプタ-発現に対する効果を調べた。IFNーα、PGE_2は、レセプタ-発現に影響を与えなかったが、IFNーγは500U/mlでFcεRIIの発現を抑制し、特異的誘導はPHA,PHA+ILー2添加で約1%,ILー4で約1〜5%となった。IFNーγによる抑制の程度も、AD患者と正常人で有為な差はなかった。以上より、AD患者と正常人で、FcεRII発現における末梢血単核球の反応性には差がないといえる。したがって、AD患者におけるFcεRII陽性細胞の高値は、各種サイトカインなどの産生の違いによる可能性が示唆された。
