1. Significance of melanin-related metabolites as biochemical markers of melanoma(1) Normal values of 5-S-cysteinyldopa(5-S-CD) and 6-hydroxy-5-methoxyindole-2-carboxylic acid(6H5MI2C) in serum and urine were obtained from 33 healthy Japanese. The urinary excretion of both markers showed significant decrease in elderly subjects as compared with middle-aged subjects, while the serum concentration showed no age-dependent differences.(2) Values of those 4 markers in melanoma patients have regularly been followed. Among the 4 markers serum 5-S-CD appears to reflect most sensitively the progression of melanoma.Some patients showed pathological values of serum 5-S-CD(above 10 nmol/L) prior to the detection by image diagnosis. On the other hand, 6H5MI2C in serum and urine was found to be of little clinical significance.(3) Serum 5-S-CD values have been analyzed every month in 2 years' period in 10 healthy subjects. No values exceeded 10 nmol/L, the upper limit of normal value, in the 240 samples.2. Improvement of HPLC determination of serum 5-S-CD The previous method for the HPLC determination of serum 5-S-CD required 80 min for each chromatographic run. Pre-treatment of serum samples has been improved to reduce the time for each analysis to 40 min, which enabled to double the analytical capacity.
1. 黑色素相关代谢物作为黑色素瘤生化标志物的意义(1)测定了33例健康日本人血清和尿液中5- s -半胱氨酸多巴(5-S-CD)和6-羟基-5-甲氧基吲哚-2-羧酸(6H5MI2C)的正常值。老年受试者尿液中两种标志物的排泄量均明显低于中年受试者,而血清浓度无年龄依赖性。(2)定期跟踪黑色素瘤患者这4项指标的值。在4种标志物中,血清5-S-CD似乎最敏感地反映黑色素瘤的进展。部分患者在影像诊断检测前血清5-S-CD已达到病理值(10 nmol/L以上)。另一方面,血清和尿液中的6H5MI2C没有什么临床意义。(3)对10例健康受试者2年内每月血清5-S-CD值进行分析。在240份样本中,没有超过正常值上限10nmol /L的。高效液相色谱法测定血清5-S-CD的改进以往的高效液相色谱法测定血清5-S-CD每次运行需要80 min。改进了血清样品的预处理,将每次分析的时间减少到40分钟,使分析能力提高了一倍。
