平成3年度に樹立した抗精液γ-GTPモノクローナル抗体を用いて、前立腺肥大・癌組織における精液γ-GTPの局在について、免疫組織化学的に検討した。材料には前立腺肥大症及び癌の前立腺生検標本及び剖検標本のホルマリン固定・パラフィン包埋切片(厚さ4μm)を用い、前立腺癌の組織分類は前立腺癌取り扱い規約に従った。免疫組織化学染色はABC法によった。一次抗体としてマウス抗精液γ-GTPモノクローナル抗体(mAb-SG1:自家製)及びウサギ抗精液γ-GTPポリクローナル抗体(PolyAb:自家製)を用いた。発色基質はジアミノベンチジンを用い、ヘマトキシリンで対比染色した。染色性により、弱陽性1+、強陽性3+、これらの中間を2+とし、更に弱陽性と陰性-が混在するものを±として、-〜3+の5段階に評価した。ちなみに正常組織では、PolyAbは肝細胞・腎尿細管上皮細胞・前立腺上皮細胞で3+を呈し、mAb-SG1は肝・腎で-、前立腺で2+を呈する。その結果、前立腺肥大ではPolyAbは2+〜3+、mAb-SG1は1+〜2+を呈した。細胞内局在には正常組織との相違は認めず、細胞質全体が染色され、特に腺腔面近くが強く染色された。前立腺肥大例は腺性過形成型よりも間質性過形成型が多く、腺上皮でのγ-GTP産生への影響は少ないものと思われた。前立腺癌ではmAb-SG1とPolyAbは同様に、分化度が低くなるほど染色性が低下する傾向を認めた。高・中分化型癌では1+〜2+が多いが、強い染色性を示す例も認められた。低分化型癌では1+以下、特に-が多く認められた。細胞内局在は、未分化細胞では全体に染色されたが、管腔を形成する癌細胞では管腔面近くが強く、細胞極性が伺われた。以上のことから、前立腺癌細胞でも精液γ-GTPが産生されているが、低分化型癌ほど免疫組織化学的に検出されなくなる傾向がある。これはPolyAbでも認められることから、エピトープの癌性変化よりも量的変化による可能性が考えられる。
