ムスカリン性アセチルコリン受容体(以下ムスカリン受容体)は、記憶のメカニズムと関係があるといわれている。痴呆の主症状は記憶の障害であり、死後脳を用いた研究から老年痴呆者の脳ではムスカリン受容体の数の減少やmRNAの増加などが報告されている。しかしDNAを直接調べた研究はない。また痴呆のもう一方の主原因である脳血管性痴呆とムスカリン受容体の関係に関する報告もみあたらない。我々の研究の目的は老年痴呆、脳血管性痴呆患者のムスカリン受容体遺伝子の塩基配列を健常者のそれと比較し、DNAレベルでの異常の有無を検討することである。〔方法〕関連施設で死後脳を収集した。半脳はホルマリン漬けにして病理学的研究に用い、残りの半脳は-80℃に保存して生化学的研究に供した。老年痴呆5例、脳血管性痴呆2例、健常コントロール1例であった。脳組織からゲノムDNAを抽出した。m1、m2に相当する部位のオリゴを合成し、PCRにて目的遺伝子を増巾した。ディデオキシ法を用いてスィークエンスを行った。〔結果および考察〕現在までに2つのグループがm1遺伝子の塩基配列を報告している。Jeffreyらは516、517番目の塩基はA、Gであり、対応するアミノ酸はスレオニン^<172>(ACA)、バリン^<173>(GTG)と報告している。一方、Peraltaらはスレオニン^<172>(ACG)、メチオニン^<173>(ATG)であったとしている。今回のわれわれの結果は、Jeffreyらの報告を支持するものであった。また、m2のスィークエンスには異常はみられなかった。

The relationship between memory and receptor is discussed in detail below. The main symptoms of dementia include impairment of memory, postmortem studies, decreased number of receptors and increased mRNA levels in Alzheimer's patients. DNA is directly modulated by DNA. The main cause of dementia is vascular dementia. The aim of our study was to compare the normal gene alignment of receptor genes in Alzheimer's disease and vascular dementia patients, and to investigate the presence or absence of DNA abnormalities.
