On the reason that little is known the properties of human liver dipeptidyl peptidase II (DPP II), we tried to determine the characteristics incliding molecular weight, kinetic parameters, amino acid sequence and cDNA structure. However, since it is not so easy to get human liver, as a preliminary experiment, we tried the purification of DPP II from human seminal plasma. The DPP II was purified to homogenity on SDS-PAGE by DEAE-Sephacel, Superdex 200 and Phenyl-Sepharose. We also determined its molecular weight (Mr), N-terminal amino acid sequence and its immunohistochemical localization in human prostatic gland. The Mr of the purified enzyme was 140,000. The enzyme preferentially clevaged the substrates such as Ala-, Phe-, Leu- and Lys-Ala-MCAs. The protein was strongly stained the luminal cells in prostatic gland. Furthermore, the amino acid sequence and cDNA structure of the enzyme indicated that DPP II in human seminal plasma is an aminopeptidase, namely alanyl aminopeptidase (AAP) … More . On the basis of our preliminary experiment using human liver and the above data, we are also planing to purify the enzyme from human liver, and to characterize under the colaboration between Japanese and Chinese research groups.It is very difficult to isolate of pathogenic bacteria because of there are many number of and many sort of mesophilic bacteria in mouth. In this study, we compared with the infection in Japanese and the infection in Chinese to establish a effective medicinal therapy.The samples were collected from the patients who were undertreating for infection in Chirlin University Hospital. The samples were cultured and tested by basteriological examination.In result, Staphylococcus, Enterococcus, Streptococcus, Peptococcus, peptostreptococcus, Corynebacterium, Lactobacillus, Norcardia, Odontomyces and Bacterionema were harvested. Susceptibility of these bacteria to some antibiotics was tested by bioassay.
