L-ascorbic acid (AsA), known as Vitamin C is a water-soluble antioxidant and an essential nutrient for the primate, because of their deficiency to biosynthesize it. Passive smoking, exposed to sidestream smoke from cigarette, is said to increase the risk of certain disease in nonsmokers, since sidestream from cigarette have much more harmful organic compounds and free radicals for body than mainstream smoke from cigarette. In this study, in order to clarify the bio-defense function of AsA against chemical stress from cigarette smoke, the AsA metabolism and the activities of drug-metabolizing enzyme were studied from the standpoint of the effect of passive smoking. Wistar male rats, which synthesized AsA,and ODS rats, which chould not synthesize AsA,were divided into two group, the control (C group) and the test (T group) groups. The animals in T group were exposed to sidestream smoke from cigarette for 2 hours everyday. After the experimental period, the AsA concentration in liver, and other tissues were measured by HPLC.When exposed to cigarette smoke, AsA levels in various tissues and activities of drug-metabolizing enzymesincreased in Wistar rats and ODS rats. It was speculated that AsA was synthesized in rats, when AsA was consumed to react with some radicals as an antioxidant or to support in raising the activities of drug-metabolizing enzymes against chemical stress. In the case of ODS rats exposed to cigarette smoke, an unexpected increase of AsA contents in various tissues was observed, and this apparent increase might be the result of some adaptation in AsA metabolism against chemical stress due to cigarette smoke. Therefore, it was concluded that the passive smoking gave a considerable effect on both AsA metabolism and activities of drug-metabolizing enzymes in rats and ODS rats.
