The purpose of this research of taking the greater part is the next thing. The way of keeping the pancreatic islets which can put up with an actual transplant is looked up by using the animal's pancreatic islets with in vitro including the pancreatic islets separate and preservation. After that, it is transplanted to the experimental diabetes model animal by using the pancreatic islets which freeze and which fused, and the effect is examined with in vivo. Furthermore, make the cause of the decline of the insuin secretion ability play due to freezing of the pancreatic islets distinct.A freezing device used the program freezer (MPF - 1000, EYELA) that a Yamaguchi University Department of Veterinary Surgery had, and liquid nitrogen. It got a pancreatic islets model from the rat, the pig which obtaines it from the center of the meat, or the cattle as well. It was observed with in vitro, the insulin secretion ability play of the pancr eatic islets was measured, and a pancreatic islets was examined in the form learning target and the function study target. With in vivo, the plan of confirming whether the diabetes condition of the recipient is monitored by each item and transplant settlement is finally extracted again and organization inspection is carried out and a recipient becomes diabetes condition again, it is progressing.Freezing of the pancr eatic islets being done if it could freeze only under the existence of dimethyl sulfoxide (DMSO) until before was done under the ethylen glycol (EG) existence. It could be proved for the first time that even EG could keep freezing of the pancreatic island in the world as a result. Moreover, EG made it clear that it was proved that cell toxicity to the pancreatic islets by soaking head time and an osmotic shock in freezing were low in comparison with DMSO and freezing could be kept more easily.
