a) Cytodifferentiation of prostatic cancer cells (Dunning prostatic adenocarcinoma) via cell to cell interaction by various fetal or neonatal mesenchymeThe neonatal rat mesenchymes (uterine and vagina) or fetal rat mesenchymes (esophagus, stomach, small intestine), or urogenital sinus mesenchymes (UGM) of fetal testicular feminization mouse were recombined with R3327 Dunning prostatic adenocarcinoma (DT). The resultant tissue recombinations were grafted under the renal capsule of male athymic mouse for 1 month. In this examination, we could observed the cytodifferentiation of the parental DT composed of small ducts lined by undifferentiated simple or multilayered squamous or cuboidal epithelial cells. Such tissue recombinations showed the enlarged ducts which were lined by low columnar epithelial cells. However, tissue recombination of rat-UGM and DT exhibited the enlarged ducts lined by tall columnar epithelial cells. This inductive difference might be caused by the number of androgen … More receptor in mesenchymal cells.From this result, we could confirm that the mesenchyme produces some cytodifferentiated factors via androgen receptor.Cytodifferentiation of human bladder cancer (T-24) via cell to cell interaction by fetal rat urogenital sinus mesenchyme (rat UGM)Human bladder cancer cells line (T-24) was recombined with rat-UGM. The resultant tissue recombination was grafted under the renal capsule of male athymic mouse for between 1and 2 months. In this examination, we could observed the cytodifferentiation of the T-24 of undifferentiated transitional cell carcinoma cells. Such tissue recombination showed the enlarged ducts which were lined by low columnar epithelial cells. Prostatic specific antigen (PSA) locally expressed on the tall columnar epithelial cells. The epithelial labeling urogenital sinus epithelium combined with rat-UGM.From this result, we could confirm that fetal mesenchyme induce the cytodifferentiation of human bladder cancer cells via epithelial-stromal interaction.
