Since the prevalence of metabolic syndrome is getting increase in Japanese, we conducted to develop an interventional program for obesity based on health education and self determination from 2000. We adopted the behavioral program to strengthen self-control of diet and exercise based on learning principles. Objectives of the program were to lose 1.0kg of weight and to reduce metabolic measurement values by 10% in dietary caloric intake and keep taking over 7,000 steps a day. Subjects were 140 residents with Body Mass Index of 24.8±3.0, aged 35-70 years in Shimane Prefecture, voluntarily participated in the present study in 2000 2002. The rate of drop out from the program for three months was 2.9%. The behavioral intervention induced significant decreases for intakes of energy, protein and fat, and an increase in the number of steps per day for participants. Since our program is based on the participants・awareness and autonomous improvement of behavior, it would be easier to continue for a long term compared to complying with specialist's complicated prescription of behavioristic modification.Trp64Arg mutation in the β_3-adrenergic receptor (β_3AR) gene is relatively common in Japanese people. However, it has not been clear whether persons with Trp64Arg mutation in the β_3AR gene tend to have obesity and difficulty in losing weight even with a restricted diet and exercise. We investigated the response of body weight and metabolic factors to behavioral intervention in Japanese women with Trp64Arg mutation in the β_3AR gene. Significant differences of anthropometric parameters were found in body weight, BMI, waist and hip circumferences and blood pressure of wild type by the intervention. However women with Trp64Arg mutation did not show significant changes in these anthropometic parameters, except for hip circumference. A significant difference was found in HDL cholesterol (HDL-C) and in the LDL-cholesterol/HDL C ratio in both genotypes. Results of the present study suggest that the Trp64Arg mutation of the β_3AR gene is associated with difficulty in losing weight through the behavioral intervention, although it is not related to obesity-related phenotypes and resting energy expenditur before the intervention.
