项目摘要

Objective: To evaluate the possibility of genetic diagnosis on drug sensitivity and malignant transformation in oral lichen planus by gene expression profiles.Methods: Six patients with oral lichen planus were included in this study, who underwent mucosal biopsies that confirmed their disease. The location of the all lesions was buccal mucosa. Clinical forms were reticular form in 4 cases and erosive form in 2 cases. We surveyed the expression of almost 16,600 genes in the 6 cases of oral lichen planus by cDNA array analysis. We extracted gene up-regulated or down-regulated in their expression with drug sensitivity. Topical corticosteroids were used in the treatment of their cases for at least four weeks.Results: Drug responders and non-responders were three cases, respectively. Out of almost 16,600 genes, expression levels of 55 genes with higher and those of 63 genes were lower in drug responders cases. But, there were not any genes to be identified significantly expressed in oral lichen planus. The lowest expression of responders was PSORS1C2, which was related to psoriasis.Conclusions: There were not significantly expressed genes in association with drug sensitivity in oral lichen planus by cDNA array analysis.
目的:探讨基因表达谱在口腔扁平苔藓药敏和恶变基因诊断中的可行性。方法:对6例口腔扁平苔藓患者进行口腔黏膜活检。病变部位均为颊粘膜。临床表现为网状4例,糜烂2例。应用基因芯片技术对6例口腔扁平苔藓组织中近16,600个基因的表达进行了检测。我们提取了对药物敏感的表达上调或下调的基因。使用局部皮质类固醇治疗至少四周。结果:药物应答者和无应答者分别为3例。在近16,600个基因中,有55个基因的表达水平较高,63个基因的表达水平较低。但是,没有发现在口腔扁平苔藓中有显著表达的基因。与银屑病相关的PSORS1C2基因表达水平最低。结论:通过基因芯片分析,口腔扁平苔藓中未发现与药物敏感相关的基因。
