ミラーシステムは観察した他者の行為を脳内で表象する働きにより、他者の行為の理解にとって重要な役割を果たすと考えられている。また、近年のfMRI研究では、ミラーシステム関連領野(下前頭回や下頭頂小葉)は、目的や意図の理解などの高次の行為の理解に関与することが報告されている。2010年度はfMRI実験により、二者の行為のインタラクションを観察・理解する際に、ミラーシステム関連領野が文脈の違いによる活動変調を示すかどうかを調べた。実験では刺激として、二人の人物のインタラクション場面を想定した映像を使用した。一方の人物は要求者であり、二つの物体のうちひとつを取って渡すように要求を出した。もう一方の人物は応答者であり、その要求に一致する物体(一致条件)もしくは不一致の物体(不一致条件)を取って要求者へ差し出した。また、要求者と応答者がともに画面上に現れる映像(二人条件)と応答者のみが現れる映像(一人条件)を用いた。実験参加者はこれらの映像を三人称視点で観察して、応答者の動作が要求に一致しているかどうかを判断した。実験の結果、左下前頭回(BA45)を含む領域で交互作用が見られた。この領域では、二人条件でのみ、一致条件に比べて不一致条件で有意な活動上昇を示した。以上の結果は、左下前頭回は観察する物体の差し出し動作自体の視覚的な情報が同一であっても文脈の違いによって活動変調を示し、この領域が社会的文脈に応じた行為の理解に重要な役割を果たすことを示唆している。
