Fulminant type 1 diabetes mellitus(FT1DM) is a subtype of T1DM, which frequently induces diabetic ketoacidosis. However, no useful marker for rapid diagnosis of FT1DM exsist. We reported that patients with FT1DM have an autoantibody against Amylaseα-2A(AMY), and developed a enzyme linked immunosorbent assay(ELISA) for detecting the antibody. Because background in the system is relatively high, so, we have improved ELISA using synthetic peptide corresponding AMY as a new antigen. When we employed peptide-7 as an antigen, we obtained good results for detecting AMY autoantibody from patients with FT1DM(sensitivity 94. 3%, specificity 93. 7% by ROC analysis), which might applicable for clinical use for rapid diagnosis of FT1DM.
暴发性1型糖尿病(FT 1DM)是T1 DM的一种亚型,常引起糖尿病酮症酸中毒。但目前尚缺乏快速诊断FT 1DM的有效指标。本文报道了FT 1DM患者存在抗淀粉酶α-2A(AMY)自身抗体,并建立了检测该抗体的酶联免疫吸附试验(ELISA)。由于该体系中本底比较高,因此,我们用合成的AMY对应肽作为新抗原,对ELISA进行了改进。当我们使用肽-7作为抗原时,我们获得了良好的结果,用于检测FT 1DM患者的AMY自身抗体(灵敏度94。3%,特异性93。ROC曲线分析表明,该方法可用于FT 1DM的快速诊断。
