Kernzytoplasmatransport von großen Proteinen und RNP-Partikeln erfordert zwei größere Komponenten: den Kernporenkomplex (NPC), eine supramolekulare Struktur, die die Zellkernhülle durchspannt, und Kerntransportrezeptoren. Letzteres ist eine Gruppe von löslichen Proteinen, die Transportsubstrate durch den NPC "eskortiert". Obwohl die Forschung in den vergangenen Jahren viele Fragen bezüglich der NPC-Struktur und der Transportrezeptoren beantwortet hat, sind die molekularen Mechanismen der Translokation der Substrat-Rezeptor-Komplexe durch den NPC selbst noch sehr unklar. Wir haben substantielle Fortschritte in der Einzelmolekülmikroskopie von zellulären Proteinen erzielt, und waren in der Lage, einen in vitro-Assay des Kerntransports, der auf isolierten Kernhüllen basiert, zu entwickeln. Wir schlagen hier vor, unsere kürzlichen Fortschritte zu kombinieren, und die Translokation einzelner Import- und Exportkomplexe durch den NPC zu studieren. Größere Einzelschritte unseres Projektes sind: 1) Lokalisierung von GFP-markierten NPC-Proteinen, die an der Translokation beteiligt sind, innerhalb des NPC; 2) Tracking von großen Importkomplexen während ihrer Translokation durch NPCs von permeabilisierten Zellen; 3) Tracking sowohl von Import- als auch Exportkomplexen durch die NPCs von isolierten Kernhüllen. Summary: Nucleocytoplasmic transport of large molecules and RNP particles involves two major components, the nuclear pore complex (NPC), a supramolecular structure spanning the nuclear envelope, and nuclear transport receptors, a family of soluble proteins escorting transport substrates through the NPC. Although much has been learnt in recent years about the NPC and transport receptors the molecular mechanisms, by which substrate-receptor complexes are translocated through the NPC, are little understood. We have achieved substantial progress in single molecule fluorescence microscopy of cellular proteins and were able to establish an in vitro assay of nuclear transport based on isolated nuclear envelopes. Here we suggest to combine our recent progress to study the translocation of single import and export complexes through the NPC. Major steps of the project are: 1) Localisation within the NPC of GFP-tagged NPC proteins implicated in translocation, 2) Tracking of large import complexes during their translocation through the NPC of permeabilized cells, 3) Tracking of both import and export complexes through the NPC of isolated nuclear envelopes. By these means we expect to resolve intermediate steps of translocation, measure transition times and generate a more detailed and quantitative model of nuclear transport.
