项目摘要

自閉症やアスペルガー症候群などの広汎性発達障害の症状の一つに、社会性の障害がある。この社会性を健全に育むには、成長過程（乳児期から青年期）の全期間で他者と触れ合うことが必要と考えられている。しかしながら、成長過程における社会性の発達時期は不明瞭なままである。そこで、本研究ではこれまでの研究成果を応用し、成長過程における社会性の発達時期を明確にすることを目的とした。さらには、幼少期の社会性が障害された後、健全な環境飼育によって社会性が回復するのかを調べ、社会性の臨界期を明らかにしようと試みた。その成果により、広汎性発達障害の理解を深めると共に、新たな予防方法・治療方法の開発への臨床的応用を目指すことができる。本研究では、マウスの生後の発達期（生後21～49日目）の間の一定期間（1週間）だけ隔離ストレス（1匹/ケージ）を負荷し、生後10週目に社会性試験等を実施して、隔離飼育期間とマウスの社会性の関連性を調べる。社会性試験には3チャンバー社会性試験装置を使用し、知らないマウスへの接近回数や周囲の滞在時間を社会性の指標として用いる予定である。マウスの成熟まで2ヶ月程度要するため、現在はマウスの体重の変化のみの結果を得ている。生後3週目、4週目、5週目の1週間だけ隔離ストレスを付加されたマウスは、隔離ストレスを付加されていないマウスと比べて、マウスの体重に明確な変化は見られなかった。この結果は、発達期の隔離飼育ストレスがマウスの食餌量やケージ内での運動量などに影響を与えていないことを示唆している。
