本研究では、ヒトの正常から病態までの衝動性の理解を意識し、時間による報酬割引に関わる分子・神経・行動の一貫した神経基盤の理解を目指す。そのため、脳内の伝達物質の一つであるセロトニンに着目し、時間による報酬割引の意思決定におけるセロトニン伝達の役割とそれに関係する神経表現を特定することを目的とする。2022年度は5-HT受容体の機能がよく知られている5-HT1A, 1B, 2A, 4受容体をPETイメージングによって受容体密度の推定を行った。その結果、5-HT1A受容体は前頭葉や海馬、扁桃体に、5-HT1B受容体は腹側淡蒼球や黒質、視覚野に、5-HT2A受容体は前頭葉や後頭葉に、5-HT4は線条体に密に存在していることがわかった。この結果はヒトの5-HT受容体サブタイプの分布と非常に類似していた。次にPETイメージングによる占有率の算出により、セロトニン伝達を阻害するための薬量を定量した。上記にあげた4種類の5-HT受容体サブタイプにおいて、約50％阻害を実現する薬量が決定された。これらの薬量を2頭のマカクザルに全身投与し報酬価値の時間割引課題を実施すると、4種類の受容体サブタイプの中で5-HT4受容体が阻害された時に行動の拒否が増加する傾向が示された。そこで報酬価値割引の数理モデルを用いて行動評価すると、時間割引係数の増加に反映されていた。行動の結果はヒトのセロトニン枯渇操作でみられる衝動性の上昇と矛盾しない結果であった。今後は例数を増やし、詳細な行動変化を確認し、結果がまとまった段階で論文化する予定である。
