项目摘要

Project Summary The goal of the Knockout Mouse Phenotyping Project (KOMP) is to provide a comprehensive catalog of mammalian gene function obtained through the creation of 8,500 gene knockouts and execution of a systematic, high-throughput phenotyping pipeline. Launched in the fall of 2011, three production/phenotyping centers including JAX were funded and assigned the task of model creation and broad-based phenotyping. The first phase succeeded in generating and phenotyping 2,500 lines. The goals of phase 2 of The Jackson Laboratory (JAX) KOMP program (KOMP2) during the current funding period (8/1/16 – 7/31/21) are to create 1000 knockout (KO) lines of mice and perform high-throughput phenotyping across organ systems using an efficient and broad testing pipeline. On March 11, 2020 the World Health Organization (WHO) declared the outbreak of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) disease (COVID-19) a world-wide pandemic. A consequence of the pandemic has been a reduction of our available personnel and services needed to reach our original goals. The purpose of this request for a funded extension beyond the current project period is to ensure continuity of well-developed model production pipelines, and to provide an uninterrupted supply of mouse lines for phenotyping. We are requesting the project be extended with funds for 12 months, with a new project period end date of 07/31/2022.
