This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Viruses in the family Reoviridae are unique molecular machines capable of transcription and post-transcription processing within the intact capsid. Cytoplasmic polyhedrosis virus (CPV), a widespread insect pathogen, belongs to the cypovirus genus in the Reoviridae family. Unlike the multiple-shelled organization of other Reoviridae viruses, CPV has a single-shelled capsid about 600 ¿ in diameter enclosing a 10-segmented dsRNA genome. Its relatively simple structural organization and unparalleled capsid stability make it an ideal system both for studying the structural basis of the endogenous RNA transcription and for pushing the resolution limit of electron cryomicroscopy reconstruction. This project is aimed at 4-5 Angstrom resolution study by single particle cryoEM with icosahedral reconstruction method.
