This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The last year's activities at the 24-ID-C insertion beamline have focused upon: 1. Crystallographic commissioning ¿ testing well characterized crystals (lysozyme, thaumatin, etc.) to fine tune the beam line and to optimize it to get the highest quality crystallographic data. 2. Further enhancement of the beamline optical and control components with the addition of new software and hardware capabilities. 3. Transition to full-time user data collection Diagnostic testing of the beamline during crystallographic commissioning has proven the capability of the 24-ID-C beam line in producing high quality crystallographic data from well diffracting crystals with low R-merge factor (~0.04 for 1.4 ¿ resolution). Additional tests have demonstrated no deterioration in data quality for exposure times as small as 100 msec. Low-energy data collection in the range of 7000-9000 eV has also shown the feasibility of solving structures by employing weak anomalous signal from sulphur atoms, reiterating the high quality of data obtainable from the 24-ID-C beam line.
