INVESTIGATION OF CLINICAL AND HORMONAL EFFECTS OF GHRP, GHRH, AND 1485 PLUS GHRH

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. Studies have shown the sustained positive effect of GHRH on growth rate and GH levels in short stature children and demonstrated a prolonged effect even after discontinuing GHRH which indicates it may be possible to restore GH secretion with GHRH. Other studies involving chronic administration of GHRH for fourteen days in elderly men showed that GHRH can reverse decreased GH and IGF levels. Several synthetic peptides, called Growth Hormone Releasing Peptides, have been developed. GHRPs have been found to release GH in a variety of animals. GHRP specifically releases GH by an action on the hypothalamus and pituitary via non-opiate, non-GHRH receptors. It is hypothesized that the activity of GHRP reflects the activity of a specific endogenous hormone and receptor. When used in combination with GHRH, GHRP very synergistically releases GH. GHRP-1 has been tested in normal men and women and revealed no laboratory or clinical adverse effects. GHRP-2 has now been given chronically to normal young men and women, elderly subjects, children and patients with pituitary tumors. In this study, the dose response effect iv, sc, and orally of 1485, another GHRP, will be studied alone and in combination with GHRH to measure efficacy and safety.
