项目摘要

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. After 20 years of work on the unnatural GHRPs, studies finally evolved into the isolation of the natural hormone designated ghrelin. Ghrelin consists of 28 amino acids including an O-n-octanolyated Ser3 residue which is essential for GH release. This hormone is the natural ligand for the growth hormone secretagogue (GHS) receptor expressed in the hypothalamus and pituitary. Ghrelin stimulates pulsatile GH release following systemic as well as intracerebroventricular administration. It is predominantly synthesized in the stomach and upper gastrointestinal tract and potentially stimulates GH as well as food intake. The specific aim of this study is to investigate the hypophysiotropic actions of ghrelin and to test its safety. It is postulated that ghrelin plays a major role in GH release and that there may be important links between the stomach, appetite and metabolism.
