This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The Mouse Research Core Facility continues to provide animal research support to the investigators of the Center for Colon Cancer Research (CCCR). The Mouse Core pays for all the cage charges incurred in maintaining all mouse colonies utilized by the target investigators, provide all equipment and reagents for PCR genotyping of mice and procurement of tissues for various studies, as well as technical support and expertise in animal procedures. In the past year, we have added the following equipment and capabilities: a BTX 830 ElectroSquare Electroporation System, which together with the isoflurane small animal anesthesia machine, provides a fully integrated system for small animal surgeries and in vivo intramuscular electroporation; and a Baker SG303 SterilGARD III laminar flow hood and a NAPCO tissue culture incubator to meet the needs of COBRE inestigators. We collaborated with the CCCR Pathology Core to collect tissues from the ApcMin/+ mouse and the Per2 ApcMin/+ mouse to carry out a pathological survey on tissues from these mice. This collaboration has resulted in a publication in the International Journal of Experimental Pathology. We also collaborated with the Histology and Imaging Core to examine the engraftment of hematopoietic stem cells (HSCs) in the small intestine and colon of the ApcMin/+ and to examine the effects of transplantation of drug resistant HSCs on tumor response to thymidylate synthase inhibitors. A manuscript is in preparation for submission to the journal Cancer Research. Several new investigators, including Lorne Hofseth (Pharmacology), Dan Dixon (Biology and South Carolina Cancer Center), and Troy Baudino (School of Medicine), have increased their use of mice. The Mouse Core provided the cage costs for these investigators, in addition to assisting them in training their personnel and procuring mouse strains for their use.
